所有樣本在 2周內的 5天時間內收集完成，其中 1個病人房間的樣本是在例行清潔前取得，另 2個病人的房間則在例行清潔後進行取樣。每天病房內會使用 5,000 ppm,的二氯異氰尿酸鈉 (Sodium dichloroisocyanurate, C3Cl2N3NaO3)兩次清潔病人的頻繁接觸區域，地板則每天使用 1,000 ppm,的二氯異氰尿酸鈉進行清潔。

研究人員用2天時間進行了空氣取樣，對於室內空氣樣本，他們使用美國SKC通用泵（37毫米濾盒和0.3微米聚四氟乙烯過濾器）以進行過濾；對於室外樣本則用德國賽多利斯Sartorius MD8微生物採樣器（帶有明膠膜過濾器）進行採集。

隨後，針對RNA依賴性RNA聚合酶和E基因，研究團隊進行了特定的實時逆轉錄酶-聚合酶鏈反應（RT-PCR），從而檢測新冠病毒的存在。

他們用循環閾值 （Cycle threshold values，Ct 值） 來量化樣本中的病毒載量，Ct 值即熒光信號達到 RT-PCR 中的閾值所需的循環數，簡單來講，Ct 值越小，表明病毒載量越高。

研究人員還收集了患者的臨床數據（癥狀、發病日期和 RT-PCR 結果）和清潔時間，並將其與採樣結果相關聯，同時如果房間環境拭子為新冠病毒陽性，團隊還測試了出現陽性的幾率。

在 15 個房間內（包括排氣扇）的取樣點中，有 13 個為病毒陽性（87％）；在 5 個衛生間（馬桶，水槽和門把手）的取樣點中有 3 個（60％）為陽性。

研究發現，未經清潔的病房馬桶、水槽、通風口等位置採集的樣本均呈新冠病毒陽性，這意味著必須十分重視個人防護、病房清潔和手部衛生。在此次新加坡團隊的研究中，雖然採集的空氣樣本仍為陰性，但是從隔離病房排氣口採集到的拭子測試呈陽性，這表明病毒仍可能隨著空氣中的氣溶膠等小液滴運動，因此嚴格地加強預防隔離病房的空氣傳染十分必要。
